Natural killer (NK) activity in children with acute lymphoblastic leukemia.
نویسنده
چکیده
The natural killer (NK) activity against K 562 target cells was explored in 20 children with acute lymphoblastic leukemia (ALL). Three of 5 patients examined at the time of diagnosis had normal NK activity. Two patients, who had more than 80% circulating blasts had significantly low NK activity. The level of NK activity of these 5 patients was inversely proportional to the percentage of blast cells contaminating the effector cell populations. After initiating induction chemotherapy, the NK activity decreased in spite of disappearance of circulating blasts. After complete remission, their NK activity returned to normal levels, and was rather higher than that of onset of the disease. NK activity of 15 children with ALL remaining in continuous complete remission was as high as that of healthy children and healthy adults in spite of intensive maintenance chemotherapy for 1/2 to 4 years. This was thought to be due to intermittent dose-schedule of the maintenance chemotherapy. 127

TRAIL-mediated killing of acute lymphoblastic leukemia by plasmacytoid dendritic cell-activated natural killer cells
Acute lymphoblastic leukemia (ALL) still frequently recurs after hematopoietic stem cell transplantation (HSCT), underscoring the need to improve the graft-versus-leukemia (GvL) effect. Natural killer (NK) cells reconstitute in the first months following HSCT when leukemia burden is at its lowest, but ALL cells have been shown to be resistant to NK cell-mediated killing. We show here that this ...
متن کاملCentral nervous system acute lymphoblastic leukemia: role of natural killer cells.
Central nervous system acute lymphoblastic leukemia (CNS-ALL) is a major clinical problem. Prophylactic therapy is neurotoxic, and a third of the relapses involve the CNS. Increased expression of interleukin 15 (IL-15) in leukemic blasts is associated with increased risk for CNS-ALL. Using in vivo models for CNS leukemia caused by mouse T-ALL and human xenografts of ALL cells, we demonstrate th...

Allotransplantation of natural killer (NK) cells has been shown to be a key factor in the control and cure of at least some hematologic diseases, such as acute myeloid leukemia or pediatric acute lymphocytic leukemia. These results support the idea that stimulation of NK cells could be an important therapeutic tool in many diseases, and several such approaches are now in clinical trials, someti...

This paper is to give a contribution to two aspects of the study of leukemia: functional characterization of human leukemia cells and cellular mechanisms leading to the killing of leukemia cells. Considering that some populations of nonmatured lymphocytic and myelolytic cells are natural killer (NK) cells or effectors of the antibody-dependent cellular cytotoxicity (K cells) , we have been look...
متن کاملExploitation of natural killer cells for the treatment of acute leukemia.
Natural killer (NK) cells play an important role in surveillance and elimination of malignant cells. Their spontaneous cytotoxicity was first demonstrated in vitro against leukemia cell lines, and NK cells might play a crucial role in the therapy of leukemia. NK cell activity is controlled by an array of germ line-encoded activating and inhibitory receptors, as well as modulating coreceptors. T...
